Evaluation of Differential Leukocyte Count by CELL-DYN 3000 Hematology Analyzer


Abstract
The CELL-DYN 3000 five-part differential was evaluated for its precision, correlation with 20 cell manual differentials, ability to process routine specimens without flagging, and ability to detect abnormalities as defined by manual differential.
A
group of 509 random patient blood samples was tested. The mean coefficients of variation for within run precision were 2.2% for the total WBC, 1.3% for neutrophils(%), 2.5% for lymphocytes(%), 2.5% for lymphocytes(%), 5.5% for monocytes(%), 20.6%
for
eosinophils(%) , and 18.8% for basophils9%). Correlation coefficients with manual differential neutrophil, lymphocyte, monocyte, eosinophil, and basophil percentages were 0.95, 0.95, 0.64, 0.85, nad 0.24, respectively. Of the 509 specimens,
107(34.38%)
were flagged. The false positive and false negative values were 8.88% and 0.83. Efficiency was calculated to be 90.29%. when combined with analysis of scattergrams, the fivepart differential count is capable of screening for abnormalities that
require
further analysis. The instrument can not be totally replaced from the manual differential count but can decrease the number of manual differential counts.
